Formulation and evaluation of chitosan based wound healing film containing croton bonplandianum extract

Wound healing is a coordinated biological process involving haemostasis and inflammation,
proliferation, and tissue remodeling. An effective wound dressing should provide a protective barrier, maintain
a suitable environment for healing, possess adequate flexibility and mechanical strength, and ideally provide
antimicrobial activity. The present study focused on the formulation and evaluation of a herbal wound-healing
film containing Croton bonplandianum leaf extract using chitosan as the main film-forming polymer. Croton
bonplandianum was selected because the submitted report describes its traditional use for cuts, wounds, skin
disorders and infections and reports wound-healing, antimicrobial, anti-inflammatory and antioxidant activities
[1,2,3,4,5,6]. The leaves were shade dried, powdered and extracted using ethanol: water (7:3). Preliminary
phytochemical screening and IR spectroscopic analysis were performed. The extract was incorporated into
chitosan/PVP films with glycerol as plasticizer by the solvent-casting method. Four formulations (F1–F4) were
prepared by varying chitosan and PVP concentrations. F4 showed the most satisfactory physical characteristics
and the highest zone of inhibition among the formulations, measuring 2.8 cm against Staphylococcus aureus,
compared with 3.0 cm for the tetracycline pellet. The optimized film showed a thickness of 0.290 mm, surface
pH 5.9, folding endurance >500 folds, 23% elongation at break and 100% flatness. The findings support the
potential of Croton bonplandianum extract in a chitosan-based herbal wound dressing, while further safety,
efficacy, release and clinical investigations are required.
